The Amyloid _ (A_) protein consists of 40-42 (43) amino acids and is cleaved from Amyloid Precursor Protein (APP) by _-secretase and subsequent _-secretase. The increased release of A_42/A_43 leads to the abnormal deposition of human A_ and the associated neurotoxicity in the brains of affected individuals.1, 2
The DRG Amyloid _ Auto-Ab ELISA Kit is a solid phase enzyme-linked immunosorbent assay (ELISA) based on the sandwich principle. The microtiter wells are coated with (1-42) _-Amyloid. An aliquot of patient sample containing A_-autoantibodies is incubated in the coated well with assay buffer. After washing a second incubation with enzyme conjugate, which is goat anti-human IgG conjugated to horseradish peroxidase, follows. After incubation the unbound conjugate is washed off.The amount of bound peroxidase is proportional to the concentration of autoantibodies against A_ in the sample. Having added the substrate solution, the intensity of colour developed is proportional to the concentration of autoantibodies against A_ in the patient sample.
